Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Reproduction of nodavirus disease was performed by experimental infection of sea bass eggs during fertilization or at larval stage 4 with 2 genetically distinguishable nodavirus strains (Sb1 and Sb2) isolated from sea bass collected along the Atlantic and Mediterranean French coast. The pathogenicity of the virus strains was assigned after detection of the virus by ELISA and immunohistochemistry (IHC). The Atlantic (Sb1) strain was more pathogenic than the Mediterranean (Sb2) strain during the fertilization step whilst both strains were pathogenic following experimental exposure of 4 d old larvae. Limited resources in the environment prevent individuals from simultaneously maximizing all life-history traits, resulting in trade-offs. The fishing of pollack which concerns essentially, France, United Kingdom and Norway, and at a lower level Ireland and Denmark, is situated between 10 and 15000 tons a year. In 1996, France was the first producer of Pollack. Since 1980, a decrease of production is observed in Europe. In France, landing reached 3000t. in 1998 (Suquet, 2001). However, the demand remaining strong, aquaculture could be a solution to satisfy the deficit between the French production and the domestic demand. |

Androgenesis is a form of uniparental reproduction leading to progenies inheriting only the paternal set of chromosomes. It has been achieved with variable success in a number of freshwater species and can be attained by artificial fertilization of genetically inactivated eggs following exposure to gamma (gamma), X-ray or UV irradiation (haploid androgenesis) and by restoration of diploidy by suppression of mitosis using a pressure or thermal shock. After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization. |
